NM_001430.5(EPAS1):c.1194G>C (p.Glu398Asp) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E398D variant (also known as c.1194G>C), located in coding exon 9 of the EPAS1 gene, results from a G to C substitution at nucleotide position 1194. The glutamic acid at codon 398 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
