Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001369.3(DNAH5):c.8722G>A (p.Glu2908Lys), citing Ambry Variant Classification Scheme 2023: The p.E2908K variant (also known as c.8722G>A), located in coding exon 52 of the DNAH5 gene, results from a G to A substitution at nucleotide position 8722. The glutamic acid at codon 2908 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
