Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001369.3(DNAH5):c.282A>G (p.Gln94=), citing Ambry Variant Classification Scheme 2023: The c.282A>G variant (also known as p.Q94Q), located in coding exon 4 of the DNAH5 gene, results from an A to G substitution at nucleotide position 282. This nucleotide substitution does not change the at codon 94. This nucleotide position is not well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration may result in the creation or strengthening of a novel splice acceptor site. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
