Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.13373C>G (p.Pro4458Arg), citing Ambry Variant Classification Scheme 2023: The p.P4458R variant (also known as c.13373C>G), located in coding exon 82 of the DNAH11 gene, results from a C to G substitution at nucleotide position 13373. The proline at codon 4458 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
