NM_001277115.2(DNAH11):c.12188T>G (p.Phe4063Cys) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.F4063C variant (also known as c.12188T>G), located in coding exon 74 of the DNAH11 gene, results from a T to G substitution at nucleotide position 12188. The phenylalanine at codon 4063 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
