NM_001232.4(CASQ2):c.919G>A (p.Asp307Asn) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D307N variant (also known as c.919G>A), located in coding exon 9 of the CASQ2 gene, results from a G to A substitution at nucleotide position 919. The aspartic acid at codon 307 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
