Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001232.4(CASQ2):c.631A>G (p.Met211Val), citing Ambry Variant Classification Scheme 2023: The p.M211V variant (also known as c.631A>G), located in coding exon 6 of the CASQ2 gene, results from an A to G substitution at nucleotide position 631. The methionine at codon 211 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
