NM_001211.6(BUB1B):c.2495G>A (p.Cys832Tyr) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C832Y variant (also known as c.2495G>A), located in coding exon 19 of the BUB1B gene, results from a G to A substitution at nucleotide position 2495. The cysteine at codon 832 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
