NM_001148.6(ANK2):c.2323C>G (p.His775Asp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H775D variant (also known as c.2323C>G), located in coding exon 21 of the ANK2 gene, results from a C to G substitution at nucleotide position 2323. The histidine at codon 775 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
