NM_001367624.2(ZNF469):c.11425G>A (p.Glu3809Lys) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: ZNF469 c.11425G>A (p.Glu3809Lys) results in a conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be tolerated. The variant allele was found at a frequency of 0.0013 in 1550462 control chromosomes, predominantly at a frequency of 0.0017 within the Non-Finnish European subpopulation in the gnomAD database, including 2 homozygotes. This frequency is not significantly higher than estimated for disease-causing variants in ZNF469, allowing no conclusion about variant significance. c.11425G>A, also known as c.11341G>A/p.E3781K, has been observed in an individual affected with keratoconus, but was also found in a healthy control (example: Lucas_2017). This report does not provide unequivocal conclusions about association of the variant with Brittle cornea syndrome 1. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 29228253). ClinVar contains an entry for this variant (Variation ID: 321026). Based on the evidence outlined above, the variant was classified as VUS-possibly benign.
