NM_000353.3(TAT):c.614T>C (p.Ile205Thr) was classified as Uncertain significance for Tyrosinemia type II by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 205 of the TAT protein (p.Ile205Thr). This variant is present in population databases (rs201857669, gnomAD 0.1%), including at least one homozygous and/or hemizygous individual. This variant has not been reported in the literature in individuals affected with TAT-related conditions. ClinVar contains an entry for this variant (Variation ID: 320410). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
