Uncertain significance — the classification assigned by Ambry Genetics to NM_006393.3(NEBL):c.2035A>G (p.Asn679Asp), citing Ambry Variant Classification Scheme 2023: The p.N679D variant (also known as c.2035A>G), located in coding exon 20 of the NEBL gene, results from an A to G substitution at nucleotide position 2035. The asparagine at codon 679 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
