Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001089.3(ABCA3):c.3796G>C (p.Glu1266Gln), citing LabCorp Variant Classification Summary - May 2015: Variant summary: ABCA3 c.3796G>C (p.Glu1266Gln) results in a conservative amino acid change located in the ABC-2 type transporter, transmembrane domain (IPR013525) of the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 251352 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.3796G>C has been reported in the literature as compound heterozygous genotype in an individual affected with Pulmonary surfactant metabolism dysfunction. These report(s) do not provide unequivocal conclusions about association of the variant with Pulmonary surfactant metabolism dysfunction. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. ClinVar contains an entry for this variant (Variation ID: 318406). Based on the evidence outlined above, the variant was classified as uncertain significance.
