This variant lies in the TSC2 gene (transcript NM_000548.5) at coding-DNA position 3733, where C is replaced by T; at the protein level this means replaces arginine at residue 1245 with tryptophan — a missense variant. Submitter rationale: The TSC2 c.3733C>T (p.R1245W) variant has not been reported in the literature to our knowledge. It was observed in 2/19924 chromosomes of the East Asian subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654), and has been reported in ClinVar (Variation ID 318328). Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
