Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001365276.2(TNXB):c.10183C>T (p.Arg3395Trp), citing LabCorp Variant Classification Summary - May 2015: Variant summary: TNXB c.10177C>T (p.Arg3393Trp) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 8.2e-06 in 244512 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.10177C>T in individuals affected with Ehlers-Danlos syndrome due to tenascin-X deficiency and no experimental evidence demonstrating its impact on protein function have been reported.
