Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000057.4(BLM):c.3418C>T (p.His1140Tyr), citing Ambry Variant Classification Scheme 2023: The p.H1140Y variant (also known as c.3418C>T), located in coding exon 17 of the BLM gene, results from a C to T substitution at nucleotide position 3418. The histidine at codon 1140 is replaced by tyrosine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
