Uncertain significance for Microphthalmia, syndromic 9 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_022369.4(STRA6):c.1957G>A (p.Val653Ile), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine with isoleucine at codon 653 of the STRA6 protein (p.Val653Ile). The valine residue is weakly conserved and there is a small physicochemical difference between valine and isoleucine. This variant is present in population databases (rs150687411, ExAC 0.03%). This variant has not been reported in the literature in individuals affected with STRA6-related conditions. ClinVar contains an entry for this variant (Variation ID: 317095). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
