Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_139057.4(ADAMTS17):c.3028G>A (p.Glu1010Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid with lysine at codon 1010 of the ADAMTS17 protein (p.Glu1010Lys). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and lysine. This variant is present in population databases (rs367651794, gnomAD 0.06%). This missense change has been observed in individual(s) with bronchopulmonary dysplasia (PMID: 31848748). ClinVar contains an entry for this variant (Variation ID: 315257). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Not Available"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Not Available"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
