Likely pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_139057.4(ADAMTS17):c.3068G>A (p.Cys1023Tyr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine, which is neutral and slightly polar, with tyrosine, which is neutral and polar, at codon 1023 of the ADAMTS17 protein (p.Cys1023Tyr). This variant is present in population databases (rs144662002, gnomAD 0.004%). This missense change has been observed in individual(s) with clinical features of ADAMTS17-related conditions (PMID: 32616716). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 315256). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change affects ADAMTS17 function (PMID: 32616716). In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic.
