Uncertain significance — the classification assigned by Ambry Genetics to NM_025233.7(COASY):c.820G>A (p.Gly274Arg), citing Ambry Variant Classification Scheme 2023: The c.907G>A (p.G303R) alteration is located in exon 4 (coding exon 3) of the COASY gene. This alteration results from a G to A substitution at nucleotide position 907, causing the glycine (G) at amino acid position 303 to be replaced by an arginine (R). Based on data from the Genome Aggregation Database (gnomAD) database, the COASY c.907G>A alteration was observed in 0.0008% (2/251326) of total alleles studied. This amino acid position is highly conserved in available vertebrate species. The p.G303R alteration is predicted to be deleterious by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
