Pathogenic for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_013275.6(ANKRD11):c.7470+2dup, citing Ambry Variant Classification Scheme 2023: The c.7470+2dupT intronic variant results from duplication of T two nucleotides after coding exon 7 of the ANKRD11 gene. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site. Based on the available evidence, this alteration is classified as pathogenic.
