Submitter rationale: The c.481_482insGACAGC (p.I161delinsRQL) alteration, located in coding exon 4 of the LDLR gene, results from an in-frame insertion of 6 nucleotides between nucleotide positions c.481 and c.482. This results in the substitution of the isoleucine residue at codon 161 for residues for arginine, glutamine, and leucine. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This alteration is predicted to be deleterious by in silico analysis (Choi, 2012).
