Likely pathogenic for Combined malonic and methylmalonic acidemia — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001243279.3(ACSF3):c.593T>G (p.Met198Arg), citing LabCorp Variant Classification Summary - May 2015: Variant summary: ACSF3 c.593T>G (p.Met198Arg) results in a non-conservative amino acid change located in the AMP-dependent synthetase/ligase domain (IPR000873) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 224322 control chromosomes (gnomAD). c.593T>G has been reported in the literature in two homozygous individuals affected with Combined Malonic And Methylmalonic Aciduria (Sloan_2011, Kuster_2018). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
