Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_005472.5(KCNE3):c.287T>G (p.Ile96Ser), citing Ambry Variant Classification Scheme 2023: The p.I96S variant (also known as c.287T>G), located in coding exon 1 of the KCNE3 gene, results from a T to G substitution at nucleotide position 287. The isoleucine at codon 96 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this variant is unclear.
