Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001845.6(COL4A1):c.82A>G (p.Lys28Glu), citing LabCorp Variant Classification Summary - May 2015: Variant summary: COL4A1 c.82A>G (p.Lys28Glu) results in a conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. This varianat has been found at a frequency of 6.794e-7 in control populations (gnomAD). To our knowledge, no occurrence of c.82A>G in individuals affected with Porencephaly 1 and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 311083). Based on the evidence outlined above, the variant was classified as uncertain significance.
