This variant lies in the VWF gene (transcript NM_000552.5) at coding-DNA position 2279, where G is replaced by A; at the protein level this means replaces arginine at residue 760 with histidine — a missense variant. Submitter rationale: Variant summary: VWF c.2279G>A (p.Arg760His) results in a non-conservative amino acid change in the encoded protein sequence near a canonical splice site. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 1.6e-05 in 251300 control chromosomes. c.2279G>A has been observed in the heterozygous state in individuals affected with type 1 Von Willebrand Disease (Riddell_2009, Liang_2017, Sadler_2021) and also together with a second pathogenic variant in individuals affected with type 2M Von Willebrand Disease (Schneppenheim_2009, Riddell_2009). These data indicate that the variant is likely to be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function and found the variant resulted in reduced expression in vitro, but did not affect collagen binding or multimer distribution (Riddell_2009).
