Pathogenic for MEGF10-related myopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001256545.2(MEGF10):c.976T>C (p.Cys326Arg), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Experimental studies have shown that this missense change affects MEGF10 function (PMID: 23954233). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. ClinVar contains an entry for this variant (Variation ID: 30966). This missense change has been observed in individual(s) with congenital myopathy (PMID: 22371254). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces cysteine, which is neutral and slightly polar, with arginine, which is basic and polar, at codon 326 of the MEGF10 protein (p.Cys326Arg).
