NM_012144.4(DNAI1):c.2041G>A (p.Glu681Lys) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E681K variant (also known as c.2041G>A), located in coding exon 20 of the DNAI1 gene, results from a G to A substitution at nucleotide position 2041. The glutamic acid at codon 681 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
