Pathogenic for Wilson disease — the classification assigned by Laboratory for Molecular Medicine, Mass General Brigham Personalized Medicine to NM_000053.4(ATP7B):c.2464dup (p.Met822fs), citing ACMG Guidelines, 2015: The p.Met822AsnfsX32 variant in ATP7B has been previously reported in a homozygous individual with Wilson disease (Gu 2003) and was absent from large population studies. This variant is predicted to cause a frameshift, which alters the protein’s amino acid sequence beginning at position 822 and leads to a premature termination codon 32 amino acids downstream. This alteration is then predicted to lead to a truncated or absent protein. Loss of function of the ATP7B gene is an established disease mechanism in autosomal recessive Wilson disease.
