This variant lies in the ATP7B gene (transcript NM_000053.4) at coding-DNA position 4098, where G is replaced by A; at the protein level this means the protein sequence is unchanged (valine at residue 1366 retained) — a synonymous variant. Submitter rationale: This synonymous variant causes a G>A nucleotide change in exon 20 of the ATP7B protein. Studies in minigene assays showed this variant causes abnormal splicing (PMID: 33719328). This variant has not been reported in individuals affected with Wilson disease in the literature. This variant has been identified in 2/246900 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
