Uncertain Significance for Lynch syndrome — the classification assigned by All of Us Research Program, National Institutes of Health to NM_000251.3(MSH2):c.*2A>C, citing ACMG Guidelines, 2015. This variant lies in the MSH2 gene (transcript NM_000251.3) at 2 bases past the stop codon (3' untranslated region), where A is replaced by C. Submitter rationale: This variant is located in the 3' untranslated region of the MSH2 gene. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with MSH2-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
