Uncertain Significance for PTEN hamartoma tumor syndrome — the classification assigned by All of Us Research Program, National Institutes of Health to NM_000314.8(PTEN):c.575C>G (p.Ala192Gly), citing ACMG Guidelines, 2015. This variant lies in the PTEN gene (transcript NM_000314.8) at coding-DNA position 575, where C is replaced by G; at the protein level this means replaces alanine at residue 192 with glycine — a missense variant. Submitter rationale: This missense variant replaces alanine with glycine at codon 192 of the PTEN protein. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with PTEN-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
