This variant lies in the RYR1 gene (transcript NM_000540.3) at coding-DNA position 56, where T is replaced by C; at the protein level this means replaces valine at residue 19 with alanine — a missense variant. Submitter rationale: This missense variant replaces valine with alanine at codon 19 of the RYR1 protein. Computational prediction suggests that this variant may have uncertain impact on protein structure and function. This variant occurs in the N-terminal region (a.a. 1 - 552) of the RYR1 protein that is considered to be a hotspot for pathogenic variants that contribute to malignant hyperthermia susceptibility (PMID: 21118704). To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with RYR1-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
