NM_000268.4(NF2):c.888T>G (p.Ile296Met) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I296M variant (also known as c.888T>G), located in coding exon 10 of the NF2 gene, results from a T to G substitution at nucleotide position 888. The isoleucine at codon 296 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
