NM_000363.5(TNNI3):c.25-1G>A was classified as Uncertain Significance for Hypertrophic cardiomyopathy by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015. This variant lies in the TNNI3 gene (transcript NM_000363.5) at the canonical splice acceptor site of the intron immediately before coding-DNA position 25, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This variant causes a G>A nucleotide substitution at the -1 position of intron 2 of the TNNI3 gene. Splice site prediction tools suggest that this variant may have a significant impact on RNA splicing. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with hypertrophic cardiomyopathy (PMID: 33241513). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Clinical relevance of loss-of-function TNNI3 splice site and truncation variants in autosomal dominant cardiovascular disorders is not clearly established. The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
