This variant lies in the WT1 gene (transcript NM_024426.6) at coding-DNA position 436, where A is replaced by C; at the protein level this means replaces lysine at residue 146 with glutamine — a missense variant. Submitter rationale: This missense variant replaces lysine with glutamine at codon 141 of the WT1 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported with a covariant WT1 c.424C>T (p.Lys142*) in an individual affected with Wilms tumor and Denys-Drash Syndrome (PMID: 28081536). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
