Uncertain Significance for Multiple endocrine neoplasia, type 1 — the classification assigned by All of Us Research Program, National Institutes of Health to NM_001370259.2(MEN1):c.-20_-14dup, citing ACMG Guidelines, 2015: This variant duplicates 7 basepairs from the 5' UTR of the MEN1 gene, located 14 basepairs upstream of the translation initiation codon. This variant does not create or introduce an alternate ATG codon nor does it affect the Kozak consensus sequence, therefore it is not predicted to impact translation initiation. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with MEN1-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
