This variant lies in the MYH7 gene (transcript NM_000257.4) at 3 bases into the intron immediately after coding-DNA position 4353, where G is replaced by A. Submitter rationale: This variant causes a G to A nucleotide substitution at the +3 position of intron 31 of the MYH7 gene. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with MYH7-related disorders in the literature. This variant has been identified in 1/251300 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Clinical relevance of loss-of-function MYH7 truncation and splice variants in autosomal dominant cardiovascular diseases is not yet clearly established. The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
