This variant lies in the ATP7B gene (transcript NM_000053.4) at coding-DNA position 3925, where G is replaced by C; at the protein level this means replaces alanine at residue 1309 with proline — a missense variant. Submitter rationale: This missense variant replaces alanine with proline at codon 1309 of the ATP7B protein. Computational prediction suggests that this variant may have deleterious impact on protein structure and function (internally defined REVEL score threshold >= 0.7, PMID: 27666373). Although functional studies have not been reported for this variant, it alters a conserved alanine residue in the phosphorylation domain of the ATP7B protein (a.a. 1004 - 1031, 1197 - 1312), a highly conserved region that is considered to be important for ATP7B protein function (PMID: 35245129; ClinVar). This variant has been reported in one individual affected with Wilson disease (PMID: 24094725). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
