Uncertain Significance for Malignant hyperthermia, susceptibility to, 5 — the classification assigned by All of Us Research Program, National Institutes of Health to NM_000069.3(CACNA1S):c.5544C>A (p.Cys1848Ter), citing ACMG Guidelines, 2015: This variant changes 1 nucleotide in exon 44/44 of the CACNA1S gene, creating a premature translation stop signal. The mutant transcript is expected to escape nonsense-mediated decay and be expressed as a truncated protein product. To our knowledge, this variant has not been reported in individuals affected with CACNA1S-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
