NM_001943.5(DSG2):c.1879+5G>C was classified as Uncertain Significance for Arrhythmogenic right ventricular cardiomyopathy by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015. This variant lies in the DSG2 gene (transcript NM_001943.5) at 5 bases into the intron immediately after coding-DNA position 1879, where G is replaced by C. Submitter rationale: This variant causes a G to C nucleotide substitution at the +5 position of intron 12 of the DSG2 gene. Splice site prediction tools predict that this variant may have a significant impact on RNA splicing. To our knowledge, RNA studies have not been reported for this variant. This variant has not been reported in individuals affected with cardiovascular disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
