This variant lies in the BRCA1 gene (transcript NM_007294.4) at coding-DNA position 526, duplicating one base; at the protein level this means shifts the reading frame starting at threonine residue 176, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This variant is predicted to result in loss of protein function through nonsense-mediated decay or protein truncation. Loss of function is an established mechanism of disease. This variant is also published as c.525insA, and it has been reported in multiple individuals and families with BRCA1-related cancers (PMID: 10880552, 12655515, 10464609). This variant is absent from large population databases, including the Genome Aggregation Database (http://gnomad.broadinstitute.org/).
