This variant lies in the LDLR gene (transcript NM_000527.5) at coding-DNA position 1093, where A is replaced by G; at the protein level this means replaces serine at residue 365 with glycine — a missense variant. Submitter rationale: This missense variant replaces serine with glycine at codon 365 of the LDLR protein. This variant is also known as p.Ser344Gly in the mature protein. This variant alters a conserved serine residue in the EGF-like repeat B of the LDLR protein (a.a. 355-393), where pathogenic missense variants are found enriched (ClinVar-LDLR). Computational prediction suggests that this variant may have a deleterious impact on protein structure and function (internally defined REVEL score threshold >= 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with familial hypercholesterolemia (PMID: 27784735). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
