NM_000069.3(CACNA1S):c.4484AGA[1] (p.Lys1496del) was classified as Uncertain Significance for Malignant hyperthermia, susceptibility to, 5 by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015: This variant results in the in-frame deletion of one amino acid in the CACNA1S protein. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in one individual in an exome sequencing study with no history of autosomal dominant malignant hyperthermia susceptability (PMID: 24195946). This variant has been identified in 1/251428 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
