Uncertain Significance for Loeys-Dietz syndrome — the classification assigned by All of Us Research Program, National Institutes of Health to NM_004612.4(TGFBR1):c.117C>T (p.His39=), citing ACMG Guidelines, 2015. This variant lies in the TGFBR1 gene (transcript NM_004612.4) at coding-DNA position 117, where C is replaced by T; at the protein level this means the protein sequence is unchanged (histidine at residue 39 retained) — a synonymous variant. Submitter rationale: This variant is located in the TGFBR1 protein. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with TGFBR1-related disorders in the literature. This variant has been identified in 2/282380 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
