Uncertain Significance for Wilson disease — the classification assigned by All of Us Research Program, National Institutes of Health to NM_000053.4(ATP7B):c.1678G>A (p.Ala560Thr), citing ACMG Guidelines, 2015. This variant lies in the ATP7B gene (transcript NM_000053.4) at coding-DNA position 1678, where G is replaced by A; at the protein level this means replaces alanine at residue 560 with threonine — a missense variant. Submitter rationale: This missense variant replaces alanine with threonine at codon 560 of the ATP7B protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with ATP7B-related disorders in the literature. This variant has been identified in 2/249538 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
