NM_024334.3(TMEM43):c.1198del (p.Glu400fs) was classified as Uncertain Significance for Arrhythmogenic right ventricular dysplasia 5 by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015: This variant deletes 1 nucleotide in exon 12 of the TMEM43 gene, causing a frameshift at the codon for the last amino acid of the protein and addition of 14 new amino acids before introducing a stop codon. This results in a protein product that is 13 amino acids longer than the normal protein product. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with TMEM43-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
