NM_000540.3(RYR1):c.5723A>G (p.Lys1908Arg) was classified as Uncertain Significance for Malignant hyperthermia, susceptibility to, 1 by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015. This variant lies in the RYR1 gene (transcript NM_000540.3) at coding-DNA position 5723, where A is replaced by G; at the protein level this means replaces lysine at residue 1908 with arginine — a missense variant. Submitter rationale: This missense variant replaces lysine with arginine at codon 1908 of the RYR1 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with autosomal dominant malignant hyperthermia susceptibility in the literature, although it is has been reported in other phenotype(s) (PMID: 33458582). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Due to insufficient evidence, this variant is classified as a Variant of Uncertain Significance for autosomal dominant malignant hyperthermia susceptibility.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
