This variant lies in the MYH7 gene (transcript NM_000257.4) at coding-DNA position 3158, where G is replaced by T; at the protein level this means replaces arginine at residue 1053 with leucine — a missense variant. Submitter rationale: This missense variant replaces arginine with leucine at codon 1053 of the MYH7 protein. Computational prediction suggests that this variant may have a deleterious impact on protein structure and function (internally defined REVEL score threshold >= 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with MYH7-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). A different variant affecting the same codon, p.Arg1053Gln, is considered to be disease-causing (ClinVar variation ID: 155814), suggesting that arginine at this position is important for MYH7 protein function. The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
