Uncertain Significance for Catecholaminergic polymorphic ventricular tachycardia — the classification assigned by All of Us Research Program, National Institutes of Health to NM_001035.3(RYR2):c.848+4A>G, citing ACMG Guidelines, 2015. This variant lies in the RYR2 gene (transcript NM_001035.3) at 4 bases into the intron immediately after coding-DNA position 848, where A is replaced by G. Submitter rationale: This variant causes an A to G nucleotide substitution at the +4 position of intron 11 of the RYR2 gene. Splice site prediction tools suggest that this variant may have a significant impact on RNA splicing. However, this prediction has not been confirmed in published RNA studies. This variant has not been reported in individuals affected with RYR2-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
